ABSTRACT
Commercially available PCR-based assays for infectious agents in semen are lacking. To understand semen burden of HIV, we developed a triple bracket- nested RT-PCR assay that is highly specific and sensitive to 20 copies of HIV RNA, and one infected cell. We have adapted this assay to detect Hepatitis C RNA, Hepatitis B DNA and CMV DNA, to sensitivities of approximately 200, 200 and 10 copies, respectively, of World Health Organization (WHO) standards.
